Abstract Severe combined immunodeficiency (SCID) arises from problems in the development of hematopoietic stem cells leading to mature T lymphocytes. Certain genotypes may affect additional lymphoid lineages. In 2014, the Primary Immune Deficiency Treatment Consortium released guidelines for diagnosing SCID, which have now been updated to include modern approaches.
