| Abstract | Helicteres isora Linn, a medicinal plant widely recognized in Ayurveda, was evaluated for its pharmacological antispasmodic potential using in vitro models of isolated guinea pig ileum. The study aimed to elucidate the plant's efficacy in reducing smooth muscle spasms, which are central to gastrointestinal discomfort and motility disorders. Extracts from the fruits of Helicteres isora were tested against standard Spasmogens such as acetylcholine, histamine, and barium chloride. The investigation revealed significant antispasmodic activity, with inhibitory effects evident on ileum contractions in response to all tested spasmogens. The mechanism is believed to involve the inhibition of acetylcholine-induced muscle contractions via modulation of muscarinic receptors, contributing to reduced muscle spasms. Comparative analysis showed the potency of the plant extract to be on par with standard antispasmodic agents like atropine and diphenhydramine hydrochloride. Acute toxicity studies demonstrated a favorable safety profile within the tested dose ranges. The results substantiate the ethno pharmacological claims regarding the use of Helicteres isora in intestinal ailments and suggest its potential for development as a natural antispasmodic therapeutic |
