Enhanced Diosgenin production in Costus speciosus via in vitro cultivation

Abstract The present study focuses on enhancing diosgenin production in Costus speciosus through optimized in vitro cultivation techniques. Diosgenin, an important steroidal precursor used in pharmaceutical industries, is traditionally obtained from field-grown plants, which show inconsistent yield due to environmental factors and long growth cycles. The research standardizes micropropagation protocols including shoot induction, shoot multiplication, root induction, and application of growth regulators to improve biomass and metabolite production. Phytochemical analysis, TLC, and HPLC studies confirmed the presence of diosgenin in both naturally grown and tissue cultured plants. HPLC quantification revealed higher diosgenin content in tissue cultured plants (0.216%) compared to naturally grown plants (0.127%). The developed protocol offers a sustainable and commercially viable alternative for large-scale diosgenin production and conservation of this medicinal species.
