Laxative Potential of the Ethanolic Leaf Extract of Ipomoea triloba (Convolvulaceae) in a Loperamide-Induced Constipation Model in Swiss Albino Mice

Abstract Constipation is a common gastrointestinal disorder characterized by infrequent bowel movements, hard stool consistency, and straining during defecation (Shi et al. 2024). Ipomoea triloba has been reported to contain flavonoids and saponins (Uka & Ukeke, 2020; StuartXchange, 2023), which are associated with potential laxative activity. This study evaluated the laxative effect of I. triloba leaf extract using Swiss albino mice. Acute oral toxicity was assessed following OECD Guideline 425, and no signs of toxicity or mortality were observed at the limit dose of 2000 mg/kg, confirming the safety of the extract. Based on this, Ipomoea triloba extract was administered at low (500 mg/kg), mid (1000 mg/kg), and high (2000 mg/kg) doses. Two experiments were conducted using twenty-five male mice each, randomly assigned into five groups: negative control (distilled water, 10 mL/kg), positive control (bisacodyl 0.25 mg/kg), and three treatment groups receiving the extract. Constipation was induced using loperamide for six days, followed by five days of treatment. Data were analyzed using one-way ANOVA followed by Tukey’s post-hoc test (p < 0.01). In the first experiment, fecal parameters showed significant dose-dependent improvement, with the most pronounced effects at mid and high doses (p < 0.001). In the second experiment, intestinal transit was significantly increased (p < 0.001), with effects comparable to the positive control and without a clear dose-dependent pattern. Overall, I. triloba exhibited significant laxative activity in loperamide-induced constipated mice.
