Abstract Papain-Like Protease (PLpro) is a key SARS-CoV-2 enzyme involved in viral replication, making it an important target for antiviral drug discovery. This in silico study compared three PLpro inhibitors—GRL-0617, VIR-250, and VIR-251. Among them, GRL-0617 showed the best binding affinity (−7.7 kcal/mol) and stable interactions, indicating its potential as a promising antiviral candidate for further studies.
