Integrated Pharmaceutical and Chromatographic Characterization of Piper longum Linn. (Pippali): Establishing a Quality Framework for Translational Investigation in Early Dyslipidemia

Abstract Background: Translation of traditional medicinal plants into reproducible research interventions requires rigorous pharmaceutical standardization and analytical characterization. Piper longum Linn. (Pippali) is an important Ayurvedic medicinal drug with a diverse phytochemical profile. Dyslipidemia is a modifiable determinant of atherosclerotic cardiovascular risk, creating interest in quality-defined botanical preparations for translational investigation.
Objective: To establish an integrated pharmaceutical, physicochemical, phytochemical and HPTLC fingerprint profile of Pippali Choornam and to define its scientific relevance as a quality-controlled preparation for future investigation in early dyslipidemia.
Methods: The investigated Pippali Choornam (Sample ID AD/24/247) underwent organoleptic and physicochemical evaluation, qualitative phytochemical screening and HPTLC fingerprinting. The HPTLC test solution was prepared by methanolic reflux extraction. Separation was performed on silica gel 60 F254 aluminium sheets using toluene:ethyl acetate:formic acid (7:3:0.1, v/v), with visualization at 254 and 366 nm and at 540 nm after derivatization.
Results: The preparation was a greenish-brown powder with characteristic odour and taste. Loss on drying was 6.53%, pH 6.05, total ash 10.03%, acid-insoluble ash 0.87%, water-soluble extractive 36.20% and alcohol-soluble extractive 25.34%. Alkaloids, glycosides, flavonoids, tannins, steroids and carbohydrates were detected. HPTLC generated nine zones at 254 nm, nine zones at 366 nm and ten zones at 540 nm after derivatization, establishing a characteristic multi-wavelength fingerprint for the investigated batch.
Conclusion: The study establishes a traceable pharmaceutical and chromatographic quality profile for the investigated Pippali Choornam. The findings provide a reproducible foundation for future marker-based, mechanistic and biological research. In view of published pharmacological interest in Piper longum constituents, standardized Pippali warrants further investigation in early dyslipidemia and lipid-associated cardiovascular-risk modulation. The present analytical study does not establish clinical cholesterol lowering or prevention of cardiovascular events.
